I'm about to go in NG+ and I want to know if killing all the NPC's (especially Fire-keepers) has any downside? Do all the NPC's respawn in NG+? Don't they?

Do enemies do more damage in NG+?

The biggest change is to enemy stats, which greatly increase with each iteration of NG+. All enemies including bosses have higher health and deal more damage. This is meant to give players a challenge, but their actual attack animations and combos stay the same.

Do all NPCs Respawn in NG+?

Kill the NPCs and loot them. They'll all respawn in NG+ and none of them will bear a grudge, so it's worth it for their armour and souls.

Do enemies give more souls in NG+?

The more powerful foes often yield up to three or four times more souls than NG, including some bosses. Souls yielded in NG+ vary from one area to another, but it's usually a multiplier of 2x, 3x or 4x compared to NG. After NG+, enemy souls and HP increase with a static percentage of the NG+ values.

Should I kill all NPCs before NG+ demon souls?

Kill off NPCs to obtain their equipments and souls. (This is not recommended for inexperienced as your Character Tendency does not reset going into NG+ and Pure Black tendency you'll get from killing NPCs comes with a number of negative effects to the user.)
